Stefano Palazzi, reduce da otto anni di lavoro al prestigioso Maudsley Hospital di Londra, ci ha segnalato la scorsa estate che il National Health Service stava affrontando il problema degli adulti con autismo, problema gravissimo, sino ad ora sommerso e che sta emergendo in tutta la sua complessità in molti paesi, Italia compresa. Fa piacere che il NHS, con la autorità che ha dentro e fuori la Gran Bretagna, affronti il problema con l’intento di dare linee guida per migliorare la situazione. Il primo atto di questa azione è stata una inchiesta on line che si è conclusa in settembre e di cui compare ora il resoconto. Sarà molto interessante leggere la strategia del DH a giorni, se intendiamo la primavera in base a ciò che dice il calendario e non a ciò che segna il termometro. Per quanto riguarda il risultato dell’inchiesta, ovvero la “Adult Autism Strategy consultation, A summary of the submissions received in response to the online consultation” gli estensori premettono, molto opportunamente, che i risultati non sono rappresentativi della generalità in quanto non è stato fatto un campionamento secondo le regole delle ricerche statistiche ma l’accesso alle risposte in Internet è stato volontario. Non ho avuto fino ad oggi la possibilità di intervenire (le cose da faresono sempre troppe rispetto al tempo ....) ma come alcuni di voi sanno nella mia ASL da circa 3 anni e mezzo stiamo portando avanti un progetto di continuità tra età evolutiva ed età adulta, che prevede un lavoro specifico con gli adulti con autismo; inoltre, nel nuovo centro per l'autismo e la sindrome di asperger nell'ospedale di Mondovì (recentemente inaugurato) abbiamo previsto uno spazio per il lavoro sulle autonomie personali e domestiche (si tratta di un mini appartamento con cucina, tavolo, divano letto e basso annesso). Abbiamo inoltre appena concluso una ricerca con l'Agenzia Regionale del Piemonte sul tema degli adulti con autismo che è stato presentato in sintesi sul Bollettino ANGSA e in un articolo nell'ultimo numero della rivista Autismo. La fatica quotidiana è però quella di far diventare nella nostra regione "sistema" (e quindi risposta organizzata sul territorio regionale) ciò che oggi sono esperienze, anche importanti, ma isolate. L'assessorato Sanità ha istuito su spinta di operatori e associazioni dei genitori un gruppo di lavoro che diventerà presto operativo (sperando che l'attuale amministrazione regionale, che ha mostrato grande attenzione, venga confermata dalle imminenti elezioni) maurizio arduino

Al momento attuale la diagnosi di Asperger viene fatta molto raramente in Italia. Vorrei essere cancellato da questa lista, anche se mi dispiace molto perdermi i comizi elettorali di Arduino (non c'è la par condicio in Piemonte?) p.s. Il sistema-Piemonte di cui parla Arduino finora ha prodotto - a Torino - un seminario con le psicologhe del Centro Autismo di Reggio Emilia (impostazione TEACCH) che doveva sfociare in un Denver Model piemontese (che non c'è mai stato). Non ho avuto fino ad oggi la possibilità di intervenire (le cose da faresono sempre troppe rispetto al tempo ....) ma come alcuni di voi sanno nella mia ASL da circa 3 anni e mezzo stiamo portando avanti un progetto di continuità tra età evolutiva ed età adulta, che prevede un lavoro specifico con gli adulti con autismo; inoltre, nel nuovo centro per l'autismo e la sindrome di asperger nell'ospedale di Mondovì (recentemente inaugurato) abbiamo previsto uno spazio per il lavoro sulle autonomie personali e domestiche (si tratta di un mini appartamento con cucina, tavolo, divano letto e basso annesso). Abbiamo inoltre appena concluso una ricerca con l'Agenzia Regionale del Piemonte sul tema degli adulti con autismo che è stato presentato in sintesi sul Bollettino ANGSA e in un articolo nell'ultimo numero della rivista Autismo. La fatica quotidiana è però quella di far diventare nella nostra regione "sistema" (e quindi risposta organizzata sul territorio regionale) ciò che oggi sono esperienze, anche importanti, ma isolate. L'assessorato Sanità ha istuito su spinta di operatori e associazioni dei genitori un gruppo di lavoro che diventerà presto operativo (sperando che l'attuale amministrazione regionale, che ha mostrato grande attenzione, venga confermata dalle imminenti elezioni)

Stefano Palazzi, reduce da otto anni di lavoro al prestigioso Maudsley Hospital di Londra, ci ha segnalato la scorsa estate che il National Health Service stava affrontando il problema degli adulti con autismo, problema gravissimo, sino ad ora sommerso e che sta emergendo in tutta la sua complessità in molti paesi, Italia compresa. Fa piacere che il NHS, con la autorità che ha dentro e fuori la Gran Bretagna, affronti il problema con l’intento di dare linee guida per migliorare la situazione. Sarà molto interessante leggere la strategia del DH a giorni, se intendiamo la primavera in base a ciò che dice il calendario e non a ciò che segna il termometro.
Per quanto riguarda il risultato dell’inchiesta, ovvero la “Adult Autism Strategy consultation, A summary of the submissions received in response to the online consultation” gli estensori premettono, molto opportunamente, che i risultati non sono rappresentativi della generalità in quanto non è stato fatto un campionamento secondo le regole delle ricerche statistiche ma l’accesso alle risposte in Internet è stato volontario. Per noi che siamo abituati alla tipologia delle persone diagnosticate autistiche in Italia, la lettura di questo documento, in cui vengono riportate molte testimonianze dirette, ci fa pensare, in alcuni momenti, che si parli di temi lontani dall’autismo, anche di quello ad alta funzione. Molte persone che si dicono appartenenti allo spettro autistico sanno esprimere per iscritto molto bene la loro situazione e i loro sentimenti e sanno navigare in Internet. Al momento attuale la diagnosi di Asperger viene fatta molto raramente in Italia. Probabilmente verrà fatta sempre di più in un prossimo futuro e pertanto molti di questi rilievi con le soluzioni suggerite possono essere utili insieme all’auspicio che le diagnosi si facciano e soprattutto si adotti una strategia per supportare chi le riceve.

Mentors for people with an ASC can be useful to help them through any difficulties that they encounter as and when they encounter them; equally, mentors, advocates or mediators may be able to help employers and colleagues understand how people with an ASC may be different to them. This again comes back to the need for employer awareness and support, which is mentioned in 69 comments.

Gentilissimi, Si ricorda che questa lista non è moderata, a differenza dell'altra, autismo e scuola, che da quest'anno è moderata e rispetta la linea politica dell'ANGSA. Chiunque vuole può farsi una lista sua, in un Paese abbastanza liberale come il nostro, e può caratterizzarla in un senso più vivace e rissoso oppure più rispettoso e sobrio, come appunto la moderazione ha realizzato selezionando gli interventi. Abbiamo avuto buoni riscontri sul nuovo corso da parte di coloro che partecipano alla lista autismo scuola, che sono molte centinaia di educatori, insegnanti e genitori, e quindi proseguiremo nella strada intrapresa. Tuttavia non crediamo sia il caso di moderare anche questa lista autismo biologia, in quanto ristretta a poco più di un centinaio di esperti, a cui sono stati ammessi soltanto alcuni laici che lo desideravano. Non possiamo certamente dare giudizi come quello di Gianni Papa (laico) su quanto viene fatto dal Dr.Arduino in Piemonte. Pertanto ANGSA se ne dissocia. Il sistema-Piemonte di cui parla Arduino finora ha prodotto - a Torino - un seminario con le psicologhe del Centro Autismo di Reggio Emilia (impostazione TEACCH) che doveva sfociare in un Denver Model piemontese (che non c'è mai stato). Spero che si faccia qualcosa di concreto, invece che proclamare miracoli e poi offrire tre ore di trattamento settimanali.

Il sistema-Piemonte di cui parla Arduino finora ha prodotto - a Torino - un seminario con le psicologhe del Centro Autismo di Reggio Emilia (impostazione TEACCH) che doveva sfociare in un Denver Model piemontese (che non c'è mai stato). Spero che si faccia qualcosa di concreto, invece che proclamare miracoli e poi offrire tre ore di trattamento settimanali.

Non ho avuto fino ad oggi la possibilità di intervenire (le cose da faresono sempre troppe rispetto al tempo ....) ma come alcuni di voi sanno nella mia ASL da circa 3 anni e mezzo stiamo portando avanti un progetto di continuità tra età evolutiva ed età adulta, che prevede un lavoro specifico con gli adulti con autismo; inoltre, nel nuovo centro per l'autismo e la sindrome di asperger nell'ospedale di Mondovì (recentemente inaugurato) abbiamo previsto uno spazio per il lavoro sulle autonomie personali e domestiche (si tratta di un mini appartamento con cucina, tavolo, divano letto e basso annesso). Abbiamo inoltre appena concluso una ricerca con l'Agenzia Regionale del Piemonte sul tema degli adulti con autismo che è stato presentato in sintesi sul Bollettino ANGSA e in un articolo nell'ultimo numero della rivista Autismo. La fatica quotidiana è però quella di far diventare nella nostra regione "sistema" (e quindi risposta organizzata sul territorio regionale) ciò che oggi sono esperienze, anche importanti, ma isolate. L'assessorato Sanità ha istuito su spinta di operatori e associazioni dei genitori un gruppo di lavoro che diventerà presto operativo (sperando che l'attuale amministrazione regionale, che ha mostrato grande attenzione, venga confermata dalle imminenti elezioni)

Sarà molto interessante leggere la strategia del DH a giorni, se intendiamo la primavera in base a ciò che dice il calendario e non a ciò che segna il termometro.
Per quanto riguarda il risultato dell’inchiesta, ovvero la “Adult Autism Strategy consultation, A summary of the submissions received in response to the online consultation” gli estensori premettono, molto opportunamente, che i risultati non sono rappresentativi della generalità in quanto non è stato fatto un campionamento secondo le regole delle ricerche statistiche ma l’accesso alle risposte in Internet è stato volontario. Per noi che siamo abituati alla tipologia delle persone diagnosticate autistiche in Italia, la lettura di questo documento, in cui vengono riportate molte testimonianze dirette, ci fa pensare, in alcuni momenti, che si parli di temi lontani dall’autismo, anche di quello ad alta funzione. Molte persone che si dicono appartenenti allo spettro autistico sanno esprimere per iscritto molto bene la loro situazione e i loro sentimenti e sanno navigare in Internet. Al momento attuale la diagnosi di Asperger viene fatta molto raramente in Italia. Probabilmente verrà fatta sempre di più in un prossimo futuro e pertanto molti di questi rilievi con le soluzioni suggerite possono essere utili insieme all’auspicio che le diagnosi si facciano e soprattutto si adotti una strategia per supportare chi le riceve.
Fatte queste premesse, molte risposte evidenziano problematiche attuali e irrisolte anche in Italia

Per noi che siamo abituati alla tipologia delle persone diagnosticate autistiche in Italia, la lettura di questo documento, in cui vengono riportate molte testimonianze dirette, ci fa pensare, in alcuni momenti, che si parli di temi lontani dall’autismo, anche di quello ad alta funzione. Molte persone che si dicono appartenenti allo spettro autistico sanno esprimere per iscritto molto bene la loro situazione e i loro sentimenti e sanno navigare in Internet. Uno di questi dice di avere difficoltà a raggiungere un gruppo di supporto perché lavora a tempo pieno, almeno così si evince dal messaggio

Al momento attuale la diagnosi di Asperger viene fatta molto raramente in Italia. Probabilmente verrà fatta sempre di più in un prossimo futuro e pertanto molti di questi rilievi con le soluzioni suggerite possono essere utili insieme all’auspicio che le diagnosi si facciano e soprattutto si adotti una strategia per supportare chi le riceve.

ho già risposto al signor Papa sul suo sito che ha continuato ad attaccare me e il sistema dei servizi piemontesi. Come ho già ripetuto in altre occasioni ciascuno è libero di criticare chi e come gli pare. Il problema è che il signor Papa non conosce nulla dell'esperienza fatta nel mio territorio in questi anni, nè penso si sia preso la briga di chiedere a chi sta sul territorio cosa si è fatto. Se vuole continuare a parlare per slogan continui a farlo. In questo modo non migliorerà certo la risposta dei servizi a favore delle persone con autismo. Ma questo non è un problema, perchè fortunatamente ci sono molte altre persone (operatori e associazioni) che agiscono invece di parlare. Per quanto riguarda i "comizi" non sono in campagna elettorale, rilevo solo che mentre qualcosa è stato fatto (ancora poco, ma sempre qualcosa è) negli ultimi 5 anni nella mia regione, nei 10 precedenti NULLA era stato fatto. maurizio arduino

Il sistema-Piemonte di cui parla Arduino finora ha prodotto - a Torino - un seminario con le psicologhe del Centro Autismo di Reggio Emilia (impostazione TEACCH) che doveva sfociare in un Denver Model piemontese (che non c'è mai stato). Spero che si faccia qualcosa di concreto, invece che proclamare miracoli e poi offrire tre ore di trattamento settimanali.

Non ho avuto fino ad oggi la possibilità di intervenire (le cose da faresono sempre troppe rispetto al tempo ....) ma come alcuni di voi sanno nella mia ASL da circa 3 anni e mezzo stiamo portando avanti un progetto di continuità tra età evolutiva ed età adulta, che prevede un lavoro specifico con gli adulti con autismo; inoltre, nel nuovo centro per l'autismo e la sindrome di asperger nell'ospedale di Mondovì (recentemente inaugurato) abbiamo previsto uno spazio per il lavoro sulle autonomie personali e domestiche (si tratta di un mini appartamento con cucina, tavolo, divano letto e basso annesso). Abbiamo inoltre appena concluso una ricerca con l'Agenzia Regionale del Piemonte sul tema degli adulti con autismo che è stato presentato in sintesi sul Bollettino ANGSA e in un articolo nell'ultimo numero della rivista Autismo. La fatica quotidiana è però quella di far diventare nella nostra regione "sistema" (e quindi risposta organizzata sul territorio regionale) ciò che oggi sono esperienze, anche importanti, ma isolate. L'assessorato Sanità ha istuito su spinta di operatori e associazioni dei genitori un gruppo di lavoro che diventerà presto operativo (sperando che l'attuale amministrazione regionale, che ha mostrato grande attenzione, venga confermata dalle imminenti elezioni)

Stefano Palazzi, reduce da otto anni di lavoro al prestigioso Maudsley Hospital di Londra, ci ha segnalato la scorsa estate che il National Health Service stava affrontando il problema degli adulti con autismo, problema gravissimo, sino ad ora sommerso e che sta emergendo in tutta la sua complessità in molti paesi, Italia compresa. Fa piacere che il NHS, con la autorità che ha dentro e fuori la Gran Bretagna, affronti il problema con l’intento di dare linee guida per migliorare la situazione. vi chiedo scusa, ma la risposta immediata da me inviata con l'ultima mail, richiede un chiarimento. Quando dico che non si è fatto nulla in Piemonte prima degli ultimi 5 anni, mi riferivo a specifici documenti della Sanità regionale in cui si parla di servizi sanitari per l'autismo. In regione di cose ne sono state fatte altre (progetti sperimentali, formazione, attività sul territorio) dalle associazioni, da vari servizi ASL e del privato sociale e, tra queste, anche una ricerca importante nel 2000, sui bisogni e i servizi, che aveva coinvolto genitori di minori e di adulti, che era stata supportata dall'allora assessorato regionale all'assistenza. Altra cosa che è giusto riconoscere alla precedente amministrazione è l'attivazione di una rete informatizzata (NPI.net) dei servizi che consente oggi di avere dati epidemiologici e consentirà di monitorare il programma regionale (e di modificare ciò che non risluterà adeguato). Come ho già ripetuto in altre occasioni ciascuno è libero di criticare chi e come gli pare. Il problema è che il signor Papa non conosce nulla dell'esperienza fatta nel mio territorio in questi anni, nè penso si sia preso la briga di chiedere a chi sta sul territorio cosa si è fatto. Se vuole continuare a parlare per slogan continui a farlo. In questo modo non migliorerà certo la risposta dei servizi a favore delle persone con autismo. Il sistema-Piemonte di cui parla Arduino finora ha prodotto - a Torino - un seminario con le psicologhe del Centro Autismo di Reggio Emilia (impostazione TEACCH) che doveva sfociare in un Denver Model piemontese (che non c'è mai stato). Spero che si faccia qualcosa di concreto, invece che proclamare miracoli e poi offrire tre ore di trattamento settimanali.

Non ho avuto fino ad oggi la possibilità di intervenire (le cose da faresono sempre troppe rispetto al tempo ....) ma come alcuni di voi sanno nella mia ASL da circa 3 anni e mezzo stiamo portando avanti un progetto di continuità tra età evolutiva ed età adulta, che prevede un lavoro specifico con gli adulti con autismo; inoltre, nel nuovo centro per l'autismo e la sindrome di asperger nell'ospedale di Mondovì (recentemente inaugurato) abbiamo previsto uno spazio per il lavoro sulle autonomie personali e domestiche (si tratta di un mini appartamento con cucina, tavolo, divano letto e basso annesso). Abbiamo inoltre appena concluso una ricerca con l'Agenzia Regionale del Piemonte sul tema degli adulti con autismo che è stato presentato in sintesi sul Bollettino ANGSA e in un articolo nell'ultimo numero della rivista Autismo. La fatica quotidiana è però quella di far diventare nella nostra regione "sistema" (e quindi risposta organizzata sul territorio regionale) ciò che oggi sono esperienze, anche importanti, ma isolate. L'assessorato Sanità ha istuito su spinta di operatori e associazioni dei genitori un gruppo di lavoro che diventerà presto operativo (sperando che l'attuale amministrazione regionale, che ha mostrato grande attenzione, venga confermata dalle imminenti elezioni)

Sarà molto interessante leggere la strategia del DH a giorni, se intendiamo la primavera in base a ciò che dice il calendario e non a ciò che segna il termometro.
Per quanto riguarda il risultato dell’inchiesta, ovvero la “Adult Autism Strategy consultation, A summary of the submissions received in response to the online consultation” gli estensori premettono, molto opportunamente, che i risultati non sono rappresentativi della generalità in quanto non è stato fatto un campionamento secondo le regole delle ricerche statistiche ma l’accesso alle risposte in Internet è stato volontario. Readers should remember that this was a qualitative consultation process, not an opinion poll; the summary or responses on the website are not representative of the population as a whole.
Per noi che siamo abituati alla tipologia delle persone diagnosticate autistiche in Italia, la lettura di questo documento, in cui vengono riportate molte testimonianze dirette, ci fa pensare, in alcuni momenti, che si parli di temi lontani dall’autismo, anche di quello ad alta funzione. Molte persone che si dicono appartenenti allo spettro autistico sanno esprimere per iscritto molto bene la loro situazione e i loro sentimenti e sanno navigare in Internet. Uno di questi dice di avere difficoltà a raggiungere un gruppo di supporto perché lavora a tempo pieno, almeno così si evince dal messaggio

Al momento attuale la diagnosi di Asperger viene fatta molto raramente in Italia. Probabilmente verrà fatta sempre di più in un prossimo futuro e pertanto molti di questi rilievi con le soluzioni suggerite possono essere utili insieme all’auspicio che le diagnosi si facciano e soprattutto si adotti una strategia per supportare chi le riceve.
Fatte queste premesse, molte risposte evidenziano problematiche attuali e irrisolte anche in Italia

Non ho avuto fino ad oggi la possibilità di intervenire (le cose da faresono sempre troppe rispetto al tempo ....) ma come alcuni di voi sanno nella mia ASL da circa 3 anni e mezzo stiamo portando avanti un progetto di continuità tra età evolutiva ed età adulta, che prevede un lavoro specifico con gli adulti con autismo; inoltre, nel nuovo centro per l'autismo e la sindrome di asperger nell'ospedale di Mondovì (recentemente inaugurato) abbiamo previsto uno spazio per il lavoro sulle autonomie personali e domestiche (si tratta di un mini appartamento con cucina, tavolo, divano letto e basso annesso). Abbiamo inoltre appena concluso una ricerca con l'Agenzia Regionale del Piemonte sul tema degli adulti con autismo che è stato presentato in sintesi sul Bollettino ANGSA e in un articolo nell'ultimo numero della rivista Autismo. La fatica quotidiana è però quella di far diventare nella nostra regione "sistema" (e quindi risposta organizzata sul territorio regionale) ciò che oggi sono esperienze, anche importanti, ma isolate. L'assessorato Sanità ha istuito su spinta di operatori e associazioni dei genitori un gruppo di lavoro che diventerà presto operativo (sperando che l'attuale amministrazione regionale, che ha mostrato grande attenzione, venga confermata dalle imminenti elezioni)

Stefano Palazzi, reduce da otto anni di lavoro al prestigioso Maudsley Hospital di Londra, ci ha segnalato la scorsa estate che il National Health Service stava affrontando il problema degli adulti con autismo, problema gravissimo, sino ad ora sommerso e che sta emergendo in tutta la sua complessità in molti paesi, Italia compresa. Sarà molto interessante leggere la strategia del DH a giorni, se intendiamo la primavera in base a ciò che dice il calendario e non a ciò che segna il termometro.
Per quanto riguarda il risultato dell’inchiesta, ovvero la “Adult Autism Strategy consultation, A summary of the submissions received in response to the online consultation” gli estensori premettono, molto opportunamente, che i risultati non sono rappresentativi della generalità in quanto non è stato fatto un campionamento secondo le regole delle ricerche statistiche ma l’accesso alle risposte in Internet è stato volontario. Per noi che siamo abituati alla tipologia delle persone diagnosticate autistiche in Italia, la lettura di questo documento, in cui vengono riportate molte testimonianze dirette, ci fa pensare, in alcuni momenti, che si parli di temi lontani dall’autismo, anche di quello ad alta funzione. Molte persone che si dicono appartenenti allo spettro autistico sanno esprimere per iscritto molto bene la loro situazione e i loro sentimenti e sanno navigare in Internet. Uno di questi dice di avere difficoltà a raggiungere un gruppo di supporto perché lavora a tempo pieno, almeno così si evince dal messaggio

Un’ altra persona che si dice affetta da ASC (Autism Spectrum Condition) dice di avere difficoltà coi trasporti pubblici e per questo usa la sua automobile e dal messaggio pare che la guidi lui.

Al momento attuale la diagnosi di Asperger viene fatta molto raramente in Italia. Probabilmente verrà fatta sempre di più in un prossimo futuro e pertanto molti di questi rilievi con le soluzioni suggerite possono essere utili insieme all’auspicio che le diagnosi si facciano e soprattutto si adotti una strategia per supportare chi le riceve.
Fatte queste premesse, molte risposte evidenziano problematiche attuali e irrisolte anche in Italia

Mentors for people with an ASC can be useful to help them through any difficulties that they encounter as and when they encounter them; equally, mentors, advocates or mediators may be able to help employers and colleagues understand how people with an ASC may be different to them. This again comes back to the need for employer awareness and support, which is mentioned in 69 comments.

The autism spectrum disorders (ASDs) are a group of conditions characterized by highly repetitive behaviour, severely restricted interests, and problems with social interactions and communication. Although ASDs are known to be highly heritable, geneticists have struggled to home in on the precise genetic elements responsible. The authors looked for copy number variants (CNVs) — segments of DNA for which the copy number differs between individual genomes — across the genomes of nearly 1,000 ASD individuals and around 1,200 healthy people. Their comparison shows that ASD sufferers carry a greater number of rare CNVs, both inherited and those that have arisen for the first time in the ASD sufferers. These variants frequently involve coding portions of the genome. The study implicates many novel genes as ASD candidates. For example, in people with an ASD, CNVs disrupt sets of genes involved in cellular proliferation, projection and motility, and GTPase/Ras signalling pathways. Cio' e' importante perche' vuol dire che studiando in modo particolare questi geni che gia' da soli scatenano la malattia si accellerera' il viaggio alla scoperta delle radici della malattia. L'autismo colpisce sei bimbi su 1000. I malati hanno difficolta' di relazione e di empatia, ovvero la capacita' di capire emozioni, stati d'animo e intenzioni altrui. La malattia e' molto complessa, ha piu' volti (ci sono diverse sfumature di gravita') e non e' riconducibile ad una sola causa, ma piu' propriamente dipende dalla complicita' di molti geni e forse anche di fattori ambientali. I ricercatori hanno cercato di ricomporre il puzzle di geni coinvolti nella genesi della malattia confrontando il genoma di 2300 persone (1000 autistici e 1300 individui sani) con una tecnica di analisi del Dna ad 'alta definizione', ovvero capace di mettere a fuoco anche minime variazioni nel Dna (una risoluzione cento volte maggiore di quella usata nel precedente lavoro del Consorzio). ''Questa volta - spiega Maestrini - grazie alle tecnologie di analisi del Dna che si fanno sempre piu' sofisticate, siamo riusciti a esaminare un milione di punti del genoma, mentre prima ne avevano esaminati solo 10 mila''. ''In particolare - precisa Maestrini - in questo studio abbiamo preso in esame le mutazioni cosiddette 'CNV', ovvero mutazioni caratterizzate dall'assenza o dalla duplicazione di piccoli pezzetti di Dna che interferiscono con la sequenza di un gene distruggendola''. Ne e' emerso un quadro davvero complesso: i CNV legati all'autismo sono molti, tutti rari, (ciascuno e' presente in meno di un individuo su 100), il che rende difficile comprendere il loro ruolo nella malattia e richiede l'analisi di un numero molto alto di pazienti. Ma la caccia ai geni dell'autismo e' tutt'altro che conclusa: ''il lavoro del consorzio va avanti - continua Maestrini - il progetto e' arruolare diverse migliaia di nuovi pazienti ed anche, quando le tecniche lo permetteranno, di arrivare a scrivere la sequenza completa del genoma di piu' pazienti''. Carissimi, Grazie Daniela per aver inoltrato il comunicato ANSA. In allegato trovate l'articolo a cui si riferisce il comunicato. Il comunicato stampa e' naturalmente un po' semplicistico, ma mi pare che il messaggio sia stato piu' o meno recepito. Sebbene le nuove conoscenze possono aiutare a migliorare i testi di diagnosi genetica, sarei pero' molto cauta riguardo alla prospettiva di test predittivi del rischio, data la complessita' della genetica dell'autismo Cerco di riassumere in altre parole il senso di questo studio: L'AGP ha studiato l'intero genoma di circa 1000 individui con autismo e 1300 individui non affetti, usando una tecnologia che permette di individuare differenze, a livello della sequenza del DNA. In particolare è stata analizzata una classe di varianti del DNA, dette copy number variants (CNVs), cioè differenze nel numero di copie di tratti di DNA (delezioni o duplicazioni), che possono influenzare l'espressione o la funzione genica in vari modi. I CNV possono essere ereditati da un genitore o insorgere come eventi de-novo, cioè nuovi. La maggior parte di essi sono variazioni comuni nel genoma di tutti gli individui, ma vari studi hanno dimostrato che alcuni CNV, rari nella popolazione, possono causare malattie genetiche. Lo studio dell'AGP mostra che individui affetti da autismo tendono ad avere un maggior numero di CNV rari che coinvolgono geni, rispetto ai controlli non affetti. Il significato funzionale della maggior parte di questi CNV è ancora da chiarire, tuttavia alcuni sono probabilmente patologici e colpiscono nuovi geni "di rischio" per l'autismo (per esempio SHANK2, SYNGAP1, DLGAP2 e PTCHD1). Si è anche cercato di identificare se i geni interrotti dai CNV svolgono funzioni cellulari comuni, ed è emerso che alcuni di questi geni hanno un ruolo al livello delle sinapsi (cioe' le connessioni tra i neuroni), altri sono implicati in meccanismi come la proliferazione e la motilità cellulare e nei sistemi di trasmissione di segnali intracellulari. Una volta identificati, questi meccanismi neuropatologici potrebbero diventare dei bersagli per lo sviluppo di nuovi trattamenti. Inoltre lo studio mostra che i CNV presenti in individui con autismo a volte coinvolgono geni che sono stati precedentemente correlati alla disabilità intellettiva, suggerendo l'esistenza di meccanismi genetici comuni. E' la scoperta messa a segno dal 'Progetto Genoma Autismo' lanciato nel 2002 dal Consorzio di studio sull'Autismo, 120 scienziati di tutto il mondo tra cui per l'Italia il gruppo di Elena Maestrini dell'Universita' di Bologna. Gli esperti hanno analizzato il Dna di migliaia di individui sani e autistici alla ricerca di geni implicati nella malattia. Annunciato sulla rivista Nature, il lavoro aiuta a capire i meccanismi molecolari nascosti dietro la malattia e in futuro potrebbe migliorare le capacita' diagnostiche e terapeutiche e magari anche portare a un test predittivo del rischio di concepire un figlio autistico. ''Tra i geni coinvolti nell'autismo trovati in questo studio - precisa Maestrini intervistata dall'ANSA - ve ne sono anche alcuni (come 'SHANK2' e 'SHANK3') che anche da soli sono sufficienti a scatenare la malattia; quindi una piccola percentuale di casi di autismo e' causata da un singolo gene difettoso''. Cio' e' importante perche' vuol dire che studiando in modo particolare questi geni che gia' da soli scatenano la malattia si accellerera' il viaggio alla scoperta delle radici della malattia. L'autismo colpisce sei bimbi su 1000. I malati hanno difficolta' di relazione e di empatia, ovvero la capacita' di capire emozioni, stati d'animo e intenzioni altrui. La malattia e' molto complessa, ha piu' volti (ci sono diverse sfumature di gravita') e non e' riconducibile ad una sola causa, ma piu' propriamente dipende dalla complicita' di molti geni e forse anche di fattori ambientali. I ricercatori hanno cercato di ricomporre il puzzle di geni coinvolti nella genesi della malattia confrontando il genoma di 2300 persone (1000 autistici e 1300 individui sani) con una tecnica di analisi del Dna ad 'alta definizione', ovvero capace di mettere a fuoco anche minime variazioni nel Dna (una risoluzione cento volte maggiore di quella usata nel precedente lavoro del Consorzio). ''Questa volta - spiega Maestrini - grazie alle tecnologie di analisi del Dna che si fanno sempre piu' sofisticate, siamo riusciti a esaminare un milione di punti del genoma, mentre prima ne avevano esaminati solo 10 mila''. ''In particolare - precisa Maestrini - in questo studio abbiamo preso in esame le mutazioni cosiddette 'CNV', ovvero mutazioni caratterizzate dall'assenza o dalla duplicazione di piccoli pezzetti di Dna che interferiscono con la sequenza di un gene distruggendola''. Ne e' emerso un quadro davvero complesso: i CNV legati all'autismo sono molti, tutti rari, (ciascuno e' presente in meno di un individuo su 100), il che rende difficile comprendere il loro ruolo nella malattia e richiede l'analisi di un numero molto alto di pazienti. Ma la caccia ai geni dell'autismo e' tutt'altro che conclusa: ''il lavoro del consorzio va avanti - continua Maestrini - il progetto e' arruolare diverse migliaia di nuovi pazienti ed anche, quando le tecniche lo permetteranno, di arrivare a scrivere la sequenza completa del genoma di piu' pazienti''. Inoltre, conclude, si studieranno i geni ora scovati per vedere il loro meccanismo d'azione e risalire ai meccanismi molecolari e cellulari dell'autismo. Intanto ci sono indizi forti del fatto che sicuramente nella malattia e' implicato un malfunzionamento delle sinapsi che interferisce con la comunicazione tra neuroni.(ANSA).
